Sequencing clinical isolates from dental plaque. Streptococcus mutans TCI-223

Plaque samples were collected from twin children with and without dental caries. S. mutans and S. sobrinus were isolated by plating plaque scrapings on selective media. Unique isolates were chosen for genome sequencing by restriction digest genotyping; libraries were constructed using the EpiBio Nextera DNA sample prep kit, and sequencing was performed using an Illumina Genome Analyzer IIx.

本研究从罹患龋齿与未罹患龋齿的双胞胎儿童中采集牙菌斑样本。通过将牙菌斑刮取物涂布于选择性培养基，分离得到变形链球菌（*Streptococcus mutans*, S. mutans）与茸毛链球菌（*Streptococcus sobrinus*, S. sobrinus）。经限制性酶切基因分型筛选出独特分离株用于基因组测序；采用EpiBio Nextera DNA样本制备试剂盒构建测序文库，并通过Illumina Genome Analyzer IIx测序平台完成测序。
