To reveal the involved gene regulatory pathways, we carried out RNA-seq transcriptomics analysis on the cells treated with or without PA, treated with PA+OA and PA+BMS. Overall design: con: HepaRG without treated for 4h. PA: HepaRG were treated with 400μM deuterium-labeled palmitate acid (PA) for 4h. PA+OA: HepaRG were treated with 400μM PA and 200μM oleic acid (OA) for 4h PA+BMS: HepaRG were treated with 50μM BMS-309403 (BMS) for 24 h during concomitant incubation with 400μM PA for 4 h.

实验设计概况:
对照组(con):未经处理的HepaRG细胞,孵育时长为4小时。

PA+OA组:将HepaRG细胞置于400μM PA与200μM油酸(OA)的混合体系中孵育4小时。
PA+BMS组:将HepaRG细胞与400μM PA共孵育4小时期间,同时用50μM BMS-309403(BMS)处理24小时。
