Explore the role of these hydrogels in wound healing, this study assessed the effects of both, Dersani Hydrogel with Alginate (DHA) and Dersani Hydrogel (DH), in human skin keratinocytes and fibroblasts gene expression profiles in a wound healing context. Sodium alginate (SA) and culture medium were also included as controls. Overall design: 1x106 HaCaT-SF or HDFn cells were seeded in 6 well plates and incubated at 37 °C until 70-80% confluence was reached. Then a gap (wound) was created by scratching the monolayer with a sterile 200 µl tip, then the cells were washed two times with phosphate buffered saline (PBS) 1X and incubated with the compounds DHA, DH and SA (0.625% w/v) dissolved in culture media containing 0.01% v/v Tween 20 during 12 h. Biological triplicates were analyzed.

本研究旨在探究此类水凝胶在伤口愈合中的作用,评估了含海藻酸盐的德尔萨尼水凝胶(Dersani Hydrogel with Alginate,DHA)与德尔萨尼水凝胶(Dersani Hydrogel,DH)在伤口愈合场景下,对人皮肤角质形成细胞和成纤维细胞基因表达谱的影响。同时以海藻酸钠(Sodium alginate,SA)与基础培养基作为对照。整体实验设计:将1×10^6个HaCaT-SF细胞或HDFn细胞接种于6孔板中,于37℃培养至细胞汇合度达70%~80%。随后使用无菌200 μl枪头刮擦细胞单层以制造人工伤口划痕,之后用1×磷酸盐缓冲液(phosphate buffered saline,PBS)洗涤细胞两次,将以含0.01% v/v吐温20(Tween 20)的培养基配制的DHA、DH与SA(0.625% w/v)分别加入体系,孵育12小时。本实验设置三次生物学重复并进行分析。
