To identify how an early life first neuroinflammatory challenge affects response to subsequent challenges. Overall design: We estabilished a neontal two-hit model: for the first hit, mice were injected subcutaneously at P3 with 5mg/kg lipopolysaccharides (L) or saline (S);for the second hit, mice received 5mg/kg LPS (L) or saline (S) intraperitoneally. Animals were divided into four groups: SS, SL, LS, LL. Mice were sacrificed at P15, 24 hours after the second hit. RNA was extracted from the hippocampus for sequencing .
为探究生命早期首次神经炎症打击对机体后续刺激应答反应的影响。
实验整体设计:本研究构建了新生小鼠双打击模型:首次打击阶段,于小鼠出生后第3天(P3)皮下注射5mg/kg脂多糖(lipopolysaccharides,简称LPS,本文中以缩写L代指)或生理盐水(本文中以缩写S代指);第二次打击阶段,通过腹腔注射给予5mg/kg LPS(即L)或生理盐水(S)。实验动物共分为四组:SS、SL、LS及LL组。于第二次打击后24小时,即小鼠出生后第15天(P15)处死所有小鼠,提取海马组织RNA进行测序。
