To evaluate the impact of IFN? secretion by influenza-specific CD8? T cells on human lung epithelial cells, we stimulated total primary human lung cells with influenza peptides in the presence or absence of an IFN?-blocking antibody. Following stimulation, epithelial cells (CD45?, CD31?, CD326?) were isolated by FACS and analyzed by single-cell RNA sequencing. Three stimulation conditions were compared including unstimulated, stimulated with influenza peptides plus isotype control antibody, and stimulated with influenza peptides plus anti-IFNg antibody. Overall design: scRNAseq of airway epithelial cells comparing three stimulation conditions. Total epithelial cells were divided into subsets based on key transcription factors post sequencing.

为评估流感特异性CD8α T细胞分泌的干扰素γ(IFNγ)对人肺上皮细胞的影响,本研究以流感肽刺激原代人肺总细胞,并设置添加或不添加干扰素γ阻断抗体的条件。刺激完成后,通过荧光激活细胞分选(FACS)分离得到上皮细胞(CD45⁻、CD31⁻、CD326⁺),随后采用单细胞RNA测序(scRNA-seq)进行分析。本研究共设置三种刺激条件:未刺激组、流感肽联合同型对照抗体刺激组、流感肽联合抗干扰素γ抗体刺激组。整体实验设计为:对气道上皮细胞开展单细胞RNA测序,对比三种刺激条件下的细胞表达差异。测序完成后,基于关键转录因子将总上皮细胞划分为不同细胞亚群。
