We provide evidence of the generation of Rdp1-mediated secondary siRNAs in vivo in fission yeast. Secondly, we show that the presence of Ago1-associated siRNAs does not guarantee robust silencing. Overall design: We obtained sequences from gel purified small RNAs of wt and rdp1delta fission yeast cells and FLAG-ago1 associated small RNAs.

本研究为裂殖酵母(fission yeast)体内Rdp1介导的次级小干扰RNA(secondary small interfering RNA, siRNA)的生成提供了实验依据。其次,本研究证实,Ago1(Argonaute 1)结合的小干扰RNA的存在并不能保证实现高效的基因沉默。整体实验设计:我们对野生型(wild type, wt)与rdp1delta裂殖酵母细胞的凝胶纯化小RNA,以及FLAG-ago1结合的小RNA进行了测序。
