Using histone H3 mutants designed so that they can only form a heterodimer, we studied the gene expression in a collection of mutants, comparing mutations to one histone H3 tail of a nucleosome compared to two histone H3 tails of a nucleosome. RNA-Seq on 13 different histone H3 mutants present in asymmetric or symmetric form, as well as 8 additional histone H3 mutants present in asymmetric form (symmetric form was presumed lethal in these remaining).
本研究利用仅能形成异二聚体(heterodimer)的组蛋白H3突变体(histone H3 mutant),对一系列突变体的基因表达谱展开研究,对比了携带单个核小体(nucleosome)组蛋白H3尾部(histone H3 tail)突变与携带两个核小体组蛋白H3尾部突变的突变体。我们对13种可分别以不对称形式(asymmetric form)或对称形式(symmetric form)存在的组蛋白H3突变体,以及另外8种仅以不对称形式存在的组蛋白H3突变体开展了RNA测序(RNA-Seq)检测;该8种额外突变体的对称形式被推测具有致死性。
