We previously identified the bHLH transcription factor SclR and found that the loss of SclR function led to significant phenotypic changes, such as rapid protein degradation and cell lysis in dextrin-polypeptone-yeast extract liquid medium. Therefore, this study presents a comparative assessment at the proteome level of the intracellular differences between an sclR-disrupted strain and a control strain using isobaric tandem mass tag (TMT) labeling for quantification.
本课题组此前已鉴定出bHLH转录因子(bHLH transcription factor)SclR,并发现SclR功能缺失会引发显著表型变化,例如在糊精-蛋白胨-酵母膏液体培养基中出现快速蛋白质降解与细胞裂解现象。为此,本研究采用同量异位素串联质量标签(isobaric tandem mass tag, TMT)标记定量技术,在蛋白质组层面对sclR敲除菌株与对照菌株的细胞内蛋白质组差异进行了比较分析。
