Metformin treatment on Fusobacterium nucleatum

High throughput RNA sequencing For RNA sequencing, F. nucleatum was incubated with 1 mM or 5 mM metformin for 7 hours, when the bacterium were under logarithmic phase. Total RNA of F. nucleatum was stabilized with RNA protect Bacteria Reagent (QIAGEN, Germany) and extracted using a QIAGEN RNeasy kit (QIAGEN, Germany) following the manufacturer’s instructions. 高通量RNA测序实验：本次实验中，将处于对数生长期的具核梭杆菌（Fusobacterium nucleatum，简称F. nucleatum）与1 mM或5 mM二甲双胍共孵育7小时。随后，采用德国QIAGEN公司的RNAprotect细菌试剂对具核梭杆菌的总RNA进行稳定保存，并依照制造商说明书使用QIAGEN RNeasy试剂盒完成总RNA提取。本实验旨在检测二甲双胍处理后具核梭杆菌的基因表达变化。
