We performed gene expression profiling on the MDA-MB-231 and MDA-MB-436 human breast cancer cell lines following siRNA-mediated inhibition of Fn14 expression as an approach to identify the mechanistic basis for Fn14 regulation of invasive capacity. Keywords: siRNA-mediated inhibition Overall design: Each cell lines was transfected with etiher an Fn14 or firefly luciferase-specific (GL2) siRNA, with untreated cells used as a reference. 2 biological replicates were performed for each array, independently grown, treated and harvested.

本研究针对MDA-MB-231与MDA-MB-436两种人类乳腺癌细胞系,在采用小干扰RNA(siRNA)介导抑制Fn14基因表达后开展基因表达谱分析,旨在解析Fn14调控细胞侵袭能力的分子机制。 关键词:siRNA介导的基因抑制 实验设计:每株细胞系均转染靶向Fn14或萤火虫荧光素酶(GL2)的siRNA,以未处理的细胞作为对照。每个基因芯片阵列设置2次生物学重复,所有样本均独立完成培养、处理与样本收获。
