Using a whole-cell proteome approach, we investigated the effect of the mug174+ gene deletion on the proteome during vegetative growth and G0/cellular quiescence in the fission yeast Schizosaccahromyces pombe. We revealed that (1) Mug174 ensured the protein expression from intron-containing genes during vegetative growth, and (2) the proteins involved in ribosome biogenesis were increased, whereas some of the G0 essential proteins were decreased in the absence of Mug174. The data from our proteome analyses indicated that Mug174 is essential for proper protein expression.
本研究采用全细胞蛋白质组学方法,探究了mug174+基因缺失对粟酒裂殖酵母(Schizosaccahromyces pombe)在营养生长及G0/细胞静止阶段蛋白质组的影响。研究结果显示:(1)Mug174可保障营养生长阶段含内含子基因的蛋白质表达;(2)在Mug174缺失的情况下,参与核糖体生物发生的蛋白质表达量升高,而部分G0期必需蛋白的表达量则降低。本蛋白质组分析数据表明,Mug174对于正常蛋白质表达至关重要。
