Gut microbiome of cold-tolerant and sensitive Piaractus mesopotamicus

We investigate the impact of low-temperature thermal stress on the gut microbiome of pacu, Piaractusmesopotamicus, an important aquaculture species indigenous to South Americas warm waters. We usedthe long-read nanopore MinION sequencer to characterize sixteen gut microbiome samples from pacuindividuals previously classified as either cold-water tolerant or sensitive. To this aim, we developed ahighly multiplexed nanopore sequencing protocol for the 16S rRNA gene based on a closed family of36mer barcodes whose specific composition allowed the minimization of crosstalk events among samples.本研究探究了低温热应激对帕库鱼（pacu，*Piaractus mesopotamicus*）肠道微生物组的影响——该鱼种是原产于南美暖水环境的重要水产养殖物种。本研究采用纳米孔长读长MinION测序仪，对16份来自帕库鱼个体的肠道微生物组样本进行解析，这些帕库鱼此前被划分为耐冷水型或冷水敏感型。为此，本研究基于一套由36碱基条形码组成的封闭家族开发了一套针对16S核糖体RNA基因的高度多重化纳米孔测序方案，该方案的条形码特定组成可最大程度降低样本间的串扰事件。
