Dendritic cells (DCs) are specialized antigen-presenting cells (APCs) linking innate and adaptive immune response. DCs are crucial for initiating adaptive immune responses for elimination of invading pathogens and also in inducing immune tolerance toward harmless components to maintain immune homeostasis. Results provide insight into the function of optineurin and its role in the maturation and function of DCs. Overall design: Bone marrow-derived dendritic cells (BMDCs) were cultured from Ctrl and DC-conditional Optn KO mice. Total RNA were obtained after LPS treatment for 8h.
树突状细胞(Dendritic cells, DCs)是一类特化的抗原呈递细胞(antigen-presenting cells, APCs),可连接固有免疫与适应性免疫应答。树突状细胞对于启动适应性免疫以清除入侵病原体,同时诱导针对无害组分的免疫耐受、维持免疫稳态至关重要。本研究结果揭示了视神经蛋白(optineurin)的功能及其在树突状细胞成熟与功能中的作用。整体实验设计:从对照组(Ctrl)与树突状细胞特异性条件性敲除optineurin(Optn)小鼠中培养骨髓来源树突状细胞(Bone marrow-derived dendritic cells, BMDCs);在脂多糖(Lipopolysaccharide, LPS)处理8小时后提取总RNA。
