To explore the underlying mechanism of STK25 in CRC, we performed single-cell transcriptome profiling of the intestinal tumor from STK25 wKO and wile-type mice.We performed dimensional reduction and unsupervised clustering of cells to identify cell groups based on their expression patterns Intestinal tumor from STK25 wKO and wile-type mice were isolated by Fluorescence-activated cell sorting(FACS) according to the presence or absence of mCherry signal and analyzed by using scRNAseq.

为探究丝氨酸/苏氨酸激酶25(STK25)在结直肠癌(CRC)中的潜在作用机制,我们对STK25敲除(wKO)及野生型(wild-type)小鼠的肠道肿瘤开展了单细胞转录组测序分析。我们基于细胞的基因表达模式,对细胞进行降维与无监督聚类分析,以鉴定不同的细胞类群。我们根据mCherry信号的有无,通过荧光激活细胞分选术(Fluorescence-activated cell sorting, FACS)分离得到STK25敲除与野生型小鼠的肠道肿瘤细胞,并采用单细胞RNA测序(scRNAseq)完成后续分析。
