本研究在Huh-7肝癌细胞系(Huh-7 hepatoma cell line)中开展全基因组CRISPR筛选(Genome-wide CRISPR screens),以鉴定调控铁死亡敏感性(ferroptosis sensitivity)的基因。具体操作:将人类GeCKO文库(Human GeCKO library)转导至Huh-7细胞,随后通过RSL-3进行筛选。实验整体设计:在经RSL-3或亚油酸(linoleic acid)联合RSL-3处理的Huh-7细胞中,采用GeCKOv2文库(GeCKOv2 library)开展全基因组CRISPR筛选。
