Protein microarray was used to identify proteins with elevated interactions with serum autoantibodies in a responding patient with rhabdomyosarcoma before and after multiple doses of HER2 CAR T cell therapy. Elevated signals were observed for multiple proteins interacting with serum autoantibodies following multiple doses of HER2-CAR T cell treatment when compared to pre-treatment serum. Overall design: Serum samples were obtained before start of treatment and subsequent follow up time points after treatment from a child with metastatic rhabdomyosarcoma enrolled in a phase 1 clinical study testing safety of HER2 targeted Chimeric Antigen Receptor (CAR) modified T cells according to Baylor College of medicine IRB approved study protocol H-24489 with the clinical trial registry number NCT00902044. The serum samples (n=6) were probed utilizing one ProtoArray™ Human Protein Microarray per sample along with a negative control.

本研究采用蛋白质微阵列(Protein microarray)技术,对1例接受多剂次HER2靶向嵌合抗原受体(Chimeric Antigen Receptor, CAR)T细胞治疗的治疗应答型横纹肌肉瘤(rhabdomyosarcoma)患者,分别采集其治疗前及多剂次治疗后的血清样本,筛选与血清自身抗体互作水平升高的蛋白质。与治疗前血清样本相比,经多剂次HER2-CAR T细胞治疗后,患者血清中与自身抗体发生互作的多种蛋白质均呈现信号上调。实验设计:招募1例转移性横纹肌肉瘤儿童受试者,其参与由贝勒医学院(Baylor College of Medicine)伦理审查委员会(Institutional Review Board, IRB)批准的I期临床试验,临床试验注册号为NCT00902044,试验方案编号为H-24489,该试验旨在评估HER2靶向嵌合抗原受体修饰T细胞的安全性。研究人员在治疗开始前及后续治疗随访时间点共采集6份血清样本,每份样本均采用1张ProtoArray™人类蛋白质微阵列(ProtoArray™ Human Protein Microarray)进行探针杂交检测,并设置阴性对照。
