The goal was to determine the chemotherapy-induced, mammalian cell death-dependent transcriptional response in a human commensal strain of E. coli. Murine intestinal epithelial cells were used to induce chemotherapy-driven cell death, and a human commensal strain of E. coli was used as the 'recipient' bacteria. Overall design: Mammalian epithelial cells were induced to die via apoptosis using the chemotherapeutic doxorubicin. Following death, cell free supernatants were used as growth medium for E. coli. Transcriptional profiles were compared to media controls and cell free supernatants where mammalian cell death had been inhibited using the pan caspase inhibitor QVD.
本研究旨在探究人类共生大肠杆菌(E. coli)菌株中,由化疗诱导且依赖哺乳动物细胞死亡的转录应答反应。实验采用小鼠肠上皮细胞诱导化疗介导的细胞死亡,并以人类共生大肠杆菌菌株作为‘受体’细菌。整体实验设计如下:使用化疗药物多柔比星(doxorubicin)诱导哺乳动物上皮细胞发生细胞凋亡。细胞死亡后,收集无细胞上清液作为大肠杆菌的生长培养基。将该培养条件下的大肠杆菌转录谱与培养基对照组,以及使用泛半胱天冬酶抑制剂QVD抑制哺乳动物细胞死亡后获得的无细胞上清液对应的转录谱进行对比。
