Assembling complete genomes for large and intricate plant species remains a formidable task, and the hexaploid wheat genome poses a particularly challenging hurdle among essential food crops. To overcome this challenge, we employed a complementary approach that integrates PacBio HiFi and ONT ultra-long (UL) read sequencing, along with crucial datasets such as chromosome conformation capture (Hi-C), Illumina, and Bionano. Our achievement presents a comprehensive telomere-to-telomere (T2T) wheat genome with no gaps and accurately includes all 21 centromeres and 42 telomeres.
组装大型复杂植物物种的完整基因组仍是一项极具挑战性的任务,六倍体小麦基因组在关键粮食作物中更是一道尤为棘手的障碍。为攻克这一难题,我们采用了整合PacBio 高保真测序(PacBio HiFi)与牛津纳米孔超长读长(ONT ultra-long, UL)测序的互补策略,并辅以染色体构象捕获(Hi-C)、Illumina测序及Bionano图谱等关键数据集。本研究成功获得了无间隙的端粒到端粒(T2T)小麦基因组,精准囊括全部21个着丝粒与42个端粒。
