This is IMAGE #1 (day 1) of a developing neuron from the optic tectum of an albino Xenopus laevis tadpole CNS, stage 47.
The cell was imaged daily over 3 days and shows the development of the dendritic arbor and axon over time.
The cell was transfected with in vivo electroporation of plasmid DNA encoding eGFP ~24 hours before the image was acquired. The intact, living tadpole was anesthetized, positioned under a glass coverslip and this image was taken directly through its transparent skin using a custom built two-photon microscope.
本图像为图像#1(第1天),来源于47期白化品系非洲爪蟾(Xenopus laevis)蝌蚪中枢神经系统(Central Nervous System, CNS)视顶盖(optic tectum)中的发育中神经元。该细胞被连续3天每日成像,完整记录了其树突分支与轴突随时间推移的发育过程。
在图像采集前约24小时,通过体内电穿孔法(in vivo electroporation)将编码增强型绿色荧光蛋白(enhanced Green Fluorescent Protein, eGFP)的质粒DNA转染入该细胞。将完整活体蝌蚪麻醉后固定于盖玻片下方,通过其透明的皮肤,使用定制双光子显微镜(two-photon microscope)直接采集得到本图像。
