We performed ChIP-seq targeting the glucocorticoid receptor (GR) in the U2OS-GR cell line and the androgen receptor (AR) in the U2OS-AR cell line. The cell lines are derived from U2OS ATTC:HTB-96 and stably transfected with an expression construct for either rat GR or human AR, respectively. The U2OS-GR cells were treated with dexamethasone (1 µM) for 90 minutes. The U2OS-AR cells were treated with R1881 (5 nM) for 4 hours.
本研究针对U2OS-GR细胞系中的糖皮质激素受体(glucocorticoid receptor, GR)以及U2OS-AR细胞系中的雄激素受体(androgen receptor, AR)开展了染色质免疫共沉淀测序(ChIP-seq)实验。这两种细胞系均源自U2OS ATTC:HTB-96细胞,分别通过稳定转染导入了大鼠GR或人AR的表达构建体。其中U2OS-GR细胞经1 μM地塞米松(dexamethasone)处理90分钟,U2OS-AR细胞经5 nM R1881处理4小时。
