The goal of this study is to unveil novel targets of the global regulator FurA using a conditional lethal mutant of Nostoc (Anabaena) sp. PCC 7120. Complete segregation of the inactivated furA gene has not been possible to date. Therefore, comparative transcriptomics of the AGcoaRFurA strain, where furA expression can be silenced and further validation with RT-PCR and EMSA assays of a set of selected genes led to identify novel FurA direct targets involved in different physiological processes.
本研究旨在利用念珠藻(Anabaena,鱼腥藻)sp. PCC 7120的条件致死突变株,挖掘全局调控因子FurA的新型靶标基因。截至目前,尚未实现失活furA基因的完全分离。因此,本研究通过可沉默furA表达的AGcoaRFurA菌株开展比较转录组学分析,并针对筛选得到的一组基因,采用逆转录聚合酶链式反应(Reverse Transcription Polymerase Chain Reaction, RT-PCR)与电泳迁移率变动分析(Electrophoretic Mobility Shift Assay, EMSA)进行验证,最终成功鉴定出参与多种生理过程的新型FurA直接靶标基因。
