The goal of this study was to compare the epigenetic landscape of perinatal hearts of cardiomyocyte-specific RXRab-deficient (edKO) and WT mice, to identify differential chromatin dynamics that are controlled by TF RXRa and RXRb. Overall design: The chromatin accessibility profiles of WT and edKO perinatal hearts mice were generated using individual P0 hearts as individual biological replicates. Transposition was performed using Tn5 enzyme (Illumina), applying size selection. Sequencing was performed in duplicate using 2×50 HiSeq 3000 (Illumina) and with an average of 25 million paired-end reads per sample.

本研究旨在比较心肌细胞特异性RXRab缺陷型(edKO)与野生型(Wild Type)小鼠围产期心脏的表观遗传景观,以鉴定受转录因子(Transcription Factor)RXRα和RXRβ调控的差异染色质动态变化。整体实验设计如下:以每只出生后0天(P0)小鼠的心脏作为独立生物学重复,制备WT与edKO小鼠围产期心脏的染色质开放谱;建库过程采用Illumina公司的Tn5转座酶进行转座反应,并进行片段大小筛选;测序环节使用Illumina HiSeq 3000平台开展2×50 bp双端测序,每个样本平均产出2500万条配对末端读段,且每个样本设置两次测序重复。
