human peripheral blood mononuclear cells (PBMCs) were transferred into immunodeficient mice to generate a mouse model with humanized T cells, and this model was applied to a stroke model. We used single-cell RNA sequencing (scRNA-seq) to reveal the heterogeneity of humanized T cells and other mouse immune cells in the humanized mouse model Overall design: immunce cells (including humanzied T cells and mouse immunce cells) of the humanized mouse were isolated by fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq.
将人外周血单个核细胞(human peripheral blood mononuclear cells,PBMCs)移植至免疫缺陷小鼠体内,构建人源化T细胞小鼠模型,并将该模型应用于脑卒中模型构建。本研究采用单细胞RNA测序(single-cell RNA sequencing,scRNA-seq)解析该人源化小鼠模型中人源化T细胞及小鼠免疫细胞的异质性。实验设计:通过荧光激活细胞分选(fluorescence-activated cell sorting,FACS)分离该人源化小鼠的免疫细胞(涵盖人源化T细胞与小鼠免疫细胞),并利用单细胞RNA测序完成相关分析。
