To dissected the roles of SMAD2/3 and SMAD4 in mouse embryonic stem cells (mESC), knockouts of SMAD2/3 (SMAD2 and SMAD3 double knockout, S2/3DKO) and SMAD4 (SMAD4 knockout, S4KO) were used to conduct RNAseq in ES and EB conditions. Overall design: WT, S2/3DKO and S4KO cells were collected at ES (D0) and embryoid body (EB) Day 4 conditions. Each sample were duplicated for RNAseq analysis.
为解析SMAD2/3与SMAD4在小鼠胚胎干细胞(mouse embryonic stem cells, mESC)中的调控作用,研究人员构建了SMAD2/3双敲除模型(SMAD2与SMAD3双敲除,S2/3DKO)与SMAD4单敲除模型(SMAD4敲除,S4KO),并分别在胚胎干细胞(ES)及拟胚体(EB)培养条件下开展RNA测序(RNAseq)分析。总体实验设计:分别在胚胎干细胞培养状态(第0天,D0)与拟胚体培养第4天(EB Day 4)的条件下,收集野生型(wild type, WT)、S2/3DKO及S4KO细胞样本,每个样本均设置2次生物学重复用于RNAseq分析。
