Pancreatic ductal adenocarcinoma (PDAC) is one of the most malignant tumors. Endoplasmic reticulum stress plays an essential role in PDAC progression. Here, we aim to identify the ERS-related genes in PDAC and build reliable risk models for diagnosis, prognosis and immunotherapy response of PDAC patients. We report HMOX1 as an important gene of ER stress related-signature we established in PDAC. The depletion of HMOX1 results in tumor growth inhibition. Our data lay the foundation for further investigating the potential mechanism. To investigate the potential mechanism of HMOX1 in the regulation of proliferation in PDAC, we conducted transcription sequencing of the AsPC-1 cells expressing RNAi against HMOX1 and scramble siRNA. We then performed gene expression profiling analysis using data obtained from RNA-seq.

胰腺导管腺癌(Pancreatic ductal adenocarcinoma, PDAC)是恶性程度最高的肿瘤之一。内质网应激(Endoplasmic reticulum stress, ERS)在PDAC的进展过程中发挥着不可或缺的作用。本研究旨在鉴定PDAC中与ERS相关的基因,并构建可靠的风险模型,以用于PDAC患者的诊断、预后评估及免疫治疗应答预测。本研究鉴定出HMOX1是我们在PDAC中构建的ERS相关特征的关键基因。敲低HMOX1可抑制肿瘤生长。本研究的数据为进一步探究其潜在作用机制奠定了基础。为了研究HMOX1调控PDAC细胞增殖的潜在机制,我们对转染靶向HMOX1的RNA干扰(RNA interference, RNAi)及阴性对照小干扰RNA(scramble siRNA)的AsPC-1细胞进行了转录测序。随后我们利用RNA测序(RNA-seq)获得的数据开展了基因表达谱分析。
