In vitro ruminal fermentation kinetics and Metagenomic characteristics of West African Fodder-tree species

DNA was extracted at the end of 96 hour in vitro incubation of fodder tree samples in sheep rumen fluid. Amplification of the V1–V2 hypervariable regions of the 16S rRNA gene was carried out using bacterial primers (27F and 357R) followed by Ion Torrent adaptors. Sequencing was done using the Ion Torrent Personal Genome Machine (PGM) system on an Ion PGM Sequencing 318™ Chip v2 BC.

将饲用树种样品在绵羊瘤胃液中进行96小时体外孵育后，于孵育结束时提取脱氧核糖核酸（DNA）。采用细菌引物（27F与357R）对16S核糖体核糖核酸（16S rRNA）基因的V1–V2高变区进行扩增，随后连接Ion Torrent测序接头。使用Ion Torrent个人基因组测序仪（PGM）系统，在Ion PGM测序318™芯片v2 BC上完成测序。
