DNA was extracted at the end of 96 hour in vitro incubation of fodder tree samples in sheep rumen fluid. Amplification of the V1âV2 hypervariable regions of the 16S rRNA gene was carried out using bacterial primers (27F and 357R) followed by Ion Torrent adaptors. Sequencing was done using the Ion Torrent Personal Genome Machine (PGM) system on an Ion PGM Sequencing 318⢠Chip v2 BC.
