Atherosclerosis involves inflammatory and thrombotic mechanisms, to which both platelets and transforming growth factor beta contribute. The effect of platelet-derived TGF beta on atherosclerosis is, however, unknown and therefore investigated. Murine platelet-selective TGF beta-deficiency (plt-TGF beta-/-) was created by a Pf4-Cre approach, and an atherosclerotic mouse model was established by functional abrogation of Ldlr and 10-15 weeks of a high-fat diet in plt-TGF beta-/- mice and their non-plt-TGF beta-/- littermates.Total RNA of the aorta was isolated according to the manufacturers instructions,the library was constructed according to the BGI Genomics.
动脉粥样硬化(Atherosclerosis)涉及炎症与血栓形成机制,血小板与转化生长因子β(transforming growth factor beta, TGF-β)均参与该病理过程。然而,血小板源性TGF-β对动脉粥样硬化的影响尚未明确,因此本研究针对该问题开展探究。研究通过Pf4-Cre重组酶系统构建了血小板选择性TGFβ缺陷小鼠(plt-TGFβ-/-);并在plt-TGFβ-/-小鼠及其非血小板选择性TGFβ缺陷同窝对照小鼠中,通过敲除低密度脂蛋白受体(Ldlr)功能并施加10~15周高脂饮食,成功建立动脉粥样硬化小鼠模型。按照制造商说明书提取主动脉总RNA,并参照华大基因(BGI Genomics)的建库流程构建测序文库。
