Fate-restricted HSCs generated distinct phenotypic MPP2. Here we report the epigenetic profiles of Vwf+Gata1+ and Vwf-Gata1+ MPP2. Overall design: MPP2 subpopulations (Vwf+Gata1+ and Vwf-Gata1+) from adult mouse bone marrow (8-12 weeks) were FACS sorted for bulk ATAC-seq (500 cells per replicate and 4 replicates per population).

命运受限的造血干细胞(Hematopoietic Stem Cells,HSCs)可产生表型独特的多能祖细胞2(Multipotent Progenitor 2,MPP2)。本研究报道了Vwf+Gata1+与Vwf-Gata1+两种MPP2的表观遗传谱。整体实验设计:从8-12周龄成年小鼠骨髓中分离得到MPP2亚群(Vwf+Gata1+与Vwf-Gata1+),经荧光激活细胞分选(Fluorescence-Activated Cell Sorting,FACS)分选后开展批量转座酶可及性测序(bulk Assay for Transposase-Accessible Chromatin using sequencing,bulk ATAC-seq),每个生物学重复样本包含500个细胞,每个亚群设置4个生物学重复。
