Purification and SmartSeq2 single cell RNA-sequencing of patient human skeletal stem cells

We have used SmartSeq2 to sequence single human skeletal stem cells (SSCs) freshly purified via FACS of tissue derived from a patient undergoing spinal fusion surgery to demonstrate this application. SmartSeq2 single cell RNA sequencing of 96 flow cytometrically purified single human skeletal stem cells from patient tissue derived during spinal fusion surgery.

本研究采用SmartSeq2技术，对通过荧光激活细胞分选（Fluorescence-Activated Cell Sorting, FACS）从脊柱融合手术患者的组织中新鲜纯化得到的人骨骼干细胞（human skeletal stem cells, SSCs）进行测序，以验证该应用的可行性。本研究针对96株经流式细胞术纯化的、源自脊柱融合手术患者组织的人骨骼干细胞，开展了SmartSeq2单细胞RNA测序。
