Mammary epithelial cell line hTert-HME1 was transfected with Control or BRCA2 siRNAs then passaged four times (12 days), then were grown in mammary epithelial growth medium with or without EGF (epithelial growth factor) for 2 passages (6 days). Overall design: Control siRNA transfection grown in media +EGF (3 replicates), control siRNA transfection grown in media -EGF (3 replicates), BRCA2 siRNA transfection grown in media +EGF (3 replicates), BRCA2 siRNA transfection grown in media -EGF (3 replicates)
将hTert-HME1人端粒酶永生化乳腺上皮细胞系分别以对照小干扰RNA(small interfering RNA,siRNA)或BRCA2靶向siRNA进行转染,随后传代培养4次(共计12天);之后将细胞置于添加或不添加表皮生长因子(epithelial growth factor,EGF)的乳腺上皮细胞生长培养基中,继续传代培养2次(共计6天)。
实验整体设计如下:设置四组实验样本,分别为对照siRNA转染且培养液添加EGF组(3个生物学重复)、对照siRNA转染且培养液不添加EGF组(3个生物学重复)、BRCA2靶向siRNA转染且培养液添加EGF组(3个生物学重复)、BRCA2靶向siRNA转染且培养液不添加EGF组(3个生物学重复)
