Raspberry root transcriptomic response to Phytophthora root rot infections and TIBA treatment

RNA-seq was used to characterise the transcriptomic response in the root tips of raspberry cv. Glen Dee (susceptible) to infection by Phytophthora rubi and treatment with an auxin transport inhibitor, TIBA. Hydroponically grown raspberry plants with established root systems were transferred to Erlenmeyer flasks containing water, with or without P. rubi inoculum and with or without TIBA. Roots were treated with 1ppm or 10ppm TIBA. After 7dpi, the root tips of three independent biological samples were collected. Differential expression analysis showed significant expression changes in roots treated with 10ppm TIBA. Analysis of differentially expressed genes identified amongst many, genes related to auxin signalling and defence response.
本研究采用RNA测序(RNA-seq)技术,对易感型树莓栽培品种Glen Dee的根尖在受树莓疫霉(Phytophthora rubi)侵染以及经生长素运输抑制剂(auxin transport inhibitor, TIBA)处理后的转录组响应进行了表征。将具有成熟根系的水培树莓植株转移至装有无菌水的锥形瓶中,设置如下处理组:分别添加或不添加树莓疫霉接种物,同时分别添加或不添加TIBA。根系分别采用1ppm与10ppm浓度的TIBA进行处理。于接种后7天(7dpi)收集3份独立生物学重复的根尖样本。差异表达分析结果显示,经10ppm TIBA处理的根系中存在显著的基因表达变化。对所鉴定出的差异表达基因进行分析后发现,在大量此类基因中,存在与生长素信号传导及防御反应相关的基因。
