Saliva of blood sucking arthropods contains a complex mixture of bioactive polypeptides that disarm their hosts blood coagulation, platelet aggregation and vasoconstriction. We here run barcoded cDNA from the salivary glands of adult female Corethrella appendiculata, a frog biting midge, and two populations of Lutzomyia umbratilis, on a single lane of a Hiseq Illumina sequencer. Sequences were assembled using Abyss and SoapdenovoTrans with several k values and the resulting contigs were further assembled by blast and cap3 iterations. There was some contamination between samples that were filtered out in the final results.
吸血节肢动物的唾液中含有复杂的生物活性多肽混合物,可拮抗宿主的血液凝固、血小板聚集与血管收缩过程。本研究利用Illumina HiSeq测序仪的单个测序通道,对成年雌性蛙叮咬蠓(Corethrella appendiculata)以及两个种群的阴蔽白蛉(Lutzomyia umbratilis)的唾液腺带条形码互补DNA(cDNA)文库进行测序。测序序列通过Abyss与SoapdenovoTrans软件,采用多种k-mer值完成组装,所得重叠群(contig)再经BLAST序列比对与Cap3迭代拼接进行进一步优化组装。样本间存在的部分交叉污染已在最终结果中予以剔除。
