We developed spinal cord organoids (SCOs) as an in vitro modeling system to help understand the pathophysiology of the spinal cord in myelomeningocele (MMC). The cell types and development of these SCOs was examined with scRNA sequencing. Overall design: SCOs were developed from human amniotic fluid induced pluripotent stem cells from both control samples and MMC samples. These SCOs were then characterized with scRNA sequencing.
本研究构建了脊髓类器官(spinal cord organoids, SCOs)体外建模体系,以助力解析脊髓脊膜膨出(myelomeningocele, MMC)中脊髓的病理生理学机制。本研究通过单细胞RNA测序(scRNA sequencing)对该类脊髓类器官的细胞类型与发育进程进行了检测分析。实验总体设计:本研究分别以对照组样本与脊髓脊膜膨出(MMC)样本来源的人类羊水诱导多能干细胞构建脊髓类器官,随后通过单细胞RNA测序对其进行表征分析。
