Analysis of sodium butyrate(NaBu) on angiotensin II(Ang II-induced cardiac fibrosis at gene expression level. The hypothesis tested in the present study was that NaBu influence the balance of gene expression of Ang II-induce cardiac fibrosis. Results provide important information of the response of NaBu to cardiac fibrosis, such as specific genes, up- or down-regulated specific cardiac cellular functions. Total RNA obtained from isolated tendon fascicles subjected to 1 or 24 hours in vitro cyclic tensile strain compared to unstrained control fascicles.

本研究在基因表达层面,探究丁酸钠(sodium butyrate,NaBu)对血管紧张素II(angiotensin II,Ang II)诱导的心肌纤维化(cardiac fibrosis)的调控作用。本研究所验证的科学假说为:丁酸钠可调控血管紧张素II诱导的心肌纤维化相关基因的表达平衡。本研究结果可为丁酸钠针对心肌纤维化的应答效应提供重要参考信息,涵盖特异性差异表达基因、心肌细胞功能的上调或下调调控等内容。实验中,将分离得到的肌腱束(tendon fascicles)分别施加1小时与24小时的体外周期性拉伸应变(in vitro cyclic tensile strain),并以未受拉伸的肌腱束作为对照,提取两组样本的总RNA(total RNA)进行后续分析。
