The aim of the study was to investigate the activation of human NK cells by IL2 through analyzing the global gene expression at different time points (0, 2, 8 and 24 hours) after culture with the cytokine IL2 at 100 IU/ml. NK cells with the CD56+/CD16+ and CD3- phenotype were negatively selected by immunomagnetic beads and re-examined by flow-cytometry to ensure greater than 90% purity . Keywords: resting and IL2 activated NK cells(time series) Overall design: RNA was extracted and pooled from 4 different donors, amplified and labled according to the manufacturer’s instruction (GeneChipU133plus2® , Affymetrix Inc, CA).

本研究旨在通过分析人类自然杀伤细胞(natural killer cells, NK)经100 IU/ml浓度的白细胞介素2(interleukin-2, IL2)细胞因子培养后不同时间点(0、2、8及24小时)的全基因表达谱,探究IL2对NK细胞的激活作用。 本研究通过免疫磁珠阴性分选获得CD56+/CD16+且CD3-表型的NK细胞,并经流式细胞术复检以确保细胞纯度不低于90%。 关键词:静息状态及IL2激活的NK细胞(时间序列) 实验整体设计:从4名不同供体中提取总RNA并混合,按照美国加利福尼亚州Affymetrix公司GeneChipU133plus2®芯片的操作指南进行RNA扩增与标记。
