We have developed self-assembled floating supported membranes which contain intergral membrane proteins such as the COVID receptor angiotensin converter enzyme 2 (ACE2). This biologically accurate and easy to assemble sample system is self-assembled onto a Oligo-Ethylene-Oxide (OEG) Self-Assembled Monolayer (SAM) coated gold surfaces using a two step self assembly process. The distance of the resulting floating protein-lipid membranes from the bulk interface can be tuned by changing the solution salt conditions. we are at a stage in this work where we are preparing for publication but require some final data sets where the compositional complexity of the floating membranes is abundantly clear. There for here we wish to undertake some final measurements using lipid and protein deutertation to enhance the clarity of the protein distributions across the membranes.
我们开发了一类自组装漂浮支撑膜体系,该体系包含新冠病毒受体血管紧张素转换酶2(ACE2)等整合膜蛋白。这一生物学特性精准且易于组装的样品体系,通过两步自组装工艺,自组装于涂覆有低聚乙二醇(OEG)自组装单分子层(SAM)的金表面。所得漂浮型蛋白脂质膜与本体界面之间的间距,可通过调整溶液盐浓度条件进行精准调控。目前本研究已进入投稿筹备阶段,我们亟需获取一批最终数据集,以充分展现漂浮膜的组成复杂性。因此,我们计划开展最终测量工作,采用脂质与蛋白质氘代技术,以提升膜上蛋白质分布的可视化清晰度。
