Transcriptional profile of human ES-cell (hPSC) derived melanoma cell models Overall design: Human ES cells were differentiated into neural crest (NC), melanoblast (MB) or melanocytes (MC). These were either WT or triple knockout (3xKO) for p53, p16, RB1. A dox-inducible cassette was integrated into the cells to activate the BRAFV600E oncogene upon addition of doxycyline.

实验设计概述:将人胚胎干细胞分化为神经嵴(neural crest,NC)、黑素母细胞(melanoblast,MB)或黑素细胞(melanocytes,MC);上述细胞均分为野生型(WT)与p53、p16、RB1三基因敲除(3xKO)两种基因型。实验中向细胞内整合了多西环素(doxycyline)诱导型基因盒,可在加入多西环素时激活BRAFV600E癌基因。
