Resident and infiltrating myeloid cells are frequently reported to be tumor-promoting upon incidence of BCBM, but these cells are often poorly discriminated. We utilized scRNAseq to transcriptionally discriminate key immune cell populations and identify that microglia are predominantly pro-inflammatory upon BCBM initiation, and driven to an antigen presenting phenotype by infiltrating T cells. Overall design: Myelinated cells were removed from whole brain homogenates of C57BL/6 and RAG1-KO mice by density gradient and sorted for CD45hi-int cells by FACS.

驻留与浸润性髓系细胞在脑转移瘤(BCBM)发生时常被报道具有促瘤活性,但这类细胞往往难以被精准区分。本研究借助单细胞RNA测序(single-cell RNA sequencing, scRNAseq)从转录组层面甄别关键免疫细胞群,并发现在BCBM发生初始阶段,小胶质细胞主要呈现促炎表型,且可被浸润性T细胞诱导为抗原呈递表型。实验整体设计:通过密度梯度离心法从C57BL/6及重组激活基因1敲除(recombination activating gene 1 knockout, RAG1-KO)小鼠的全脑匀浆中去除髓鞘化细胞,再经荧光激活细胞分选(fluorescence-activated cell sorting, FACS)分选出CD45高-中间表达水平的细胞。
