Esophageal cancer presents significant challenges due to limited treatment options and poor prognosis, particularly in advanced stages. Dysregulated long non-coding RNAs (lncRNAs) are implicated in cancer progression and treatment resistance. This study investigated the roles of dysregulated lncRNA, identified through lncRNA-seq, and its downstream target gene in esophageal squamous cell carcinoma(ESCC). In order to investigate the molecular mechanism of different sensitivity of esophageal squamous cell carcinoma to chemotherapy, we performed incrNA-seg to detect the differentially expressed IncRNA in esophageal squamous cell carcinoma tissues between chemotherapy-sensitive and chemotherapy-resistant groups. To comprehensively explore LncRNA and its downstream molecules in ESCC, cell functions including cell viability, cell proliferation and flow cytometry analysis were examined after interference and overexpression of differential IncRNA.

食管癌因治疗手段有限、预后不佳,尤其在晚期阶段面临显著诊疗挑战。失调的长链非编码RNA(long non-coding RNAs,lncRNAs)已被证实与肿瘤进展及治疗耐药密切相关。本研究针对通过长链非编码RNA测序(lncRNA-seq)筛选得到的失调长链非编码RNA及其下游靶基因,探讨其在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)中的作用。为探究食管鳞状细胞癌对化疗敏感性差异的分子机制,本研究通过长链非编码RNA测序(lncRNA-seq),检测化疗敏感组与化疗耐药组食管鳞状细胞癌组织中差异表达的长链非编码RNA。为全面解析食管鳞状细胞癌中长链非编码RNA及其下游分子的调控机制,本研究在对差异表达长链非编码RNA进行干扰及过表达处理后,检测了细胞活力、细胞增殖能力及流式细胞术分析等多项细胞功能学指标。
