Xrn1 ko A549 cells were infected with WT or PR8-PA(∆X), or mock infected. RNA was extracted to run 5’ RACE using primers ~150-200 nt downstream of GCUG sites that are predicted to be inside a hairpin loop within viral mRNAs (in segment 4 or 8 as indicated)
将Xrn1基因敲除的A549细胞分别以野生型（WT）、PR8-PA(∆X)毒株感染，同时设置空白感染对照。提取细胞总RNA后，设计针对被预测存在于病毒mRNA发夹环结构内的GCUG位点下游约150~200 nt区域的引物，开展5' 快速扩增cDNA末端（5’ RACE）实验，上述GCUG位点对应第4或第8基因组节段（如标注所示）
