We introduce in vivo mRNA display, a technology that converts a variety of proteomics applications into a DNA sequencing problem. Overall design: In vivo expressed proteins are coupled with their encoding mRNAs via a high-affinity stem-loop RNA binding domain interaction, enabling high-throughput identification of proteins with high sensitivity and specificity by next-generation DNA sequencing.

我们提出了体内mRNA展示(in vivo mRNA display)技术,该技术可将各类蛋白质组学(proteomics)应用转化为DNA测序问题。整体实验设计:体内表达的蛋白质通过高亲和力茎环RNA结合结构域的相互作用,与其编码的mRNA相偶联,从而能够借助下一代DNA测序(next-generation DNA sequencing)技术,实现高灵敏度、高特异性的蛋白质高通量鉴定。
