KW-2449 and VPA exert therapeutic effects on human neurons and cerebral organoids derived from MECP2-null hESCs

To gain a deeper insight into roles of MECP2 in hESCs derived organoids, we performed RNA-seq to analyze the transcriptome changes after MECP2 deletion in hESCs-derived organoids. Total RNA was extracted from WT and MECP2-KO organoids treated after KW-2449 or VPA,and generated by deep sequencing, in two biological replicates, using novaseq 6000.
为深入解析MECP2在人类胚胎干细胞(hESCs)来源类器官中的功能,我们通过RNA测序(RNA-seq)分析了MECP2缺失的hESCs来源类器官的转录组变化。实验从经KW-2449或丙戊酸钠(VPA)处理的野生型(WT)与MECP2基因敲除(MECP2-KO)类器官中提取总RNA,并采用NovaSeq 6000测序平台完成了两个生物学重复的深度测序。
