We performed methylated RNA immunoprecipitation followed by sequencing (MeRIP–seq) combined with transcription sequencing to clarify the mechanism underlined YTHDF3 in CRC More than 50 ug of total RNA is sufficient for followed RNA fragmentation and immunoprecipitation according to the manuscript of the Magna MeRIP™ m6A Kit (Merck, Darmstadt, Germany).
本研究采用甲基化RNA免疫沉淀测序(MeRIP–seq,methylated RNA immunoprecipitation followed by sequencing)联合转录组测序的实验策略,以阐明YTHDF3在结直肠癌(Colorectal Cancer, CRC)中的作用机制。依据德国达姆施塔特默克(Merck)公司Magna MeRIP™ m6A试剂盒的官方操作手册,仅需50 μg以上的总RNA即可开展后续的RNA片段化与免疫沉淀实验。
