We performed single cell transcriptional profiling of mouse corneal epithelium in a mouse model of dry eye disease. Overall design: We prepared cell suspensions of corneal epithelium: one from control and one from dry eye disease (n = 7 male C57BL/6J mice, 10 weeks old). We excised the right lacrimal glands (both extra- and intra-orbital) following previously described protocols (Shinomiya et al., 2018). We sacrificed mice 9 days after lacrimal gland excision. Left control eyes were pooled (n = 7 eyes), and right dry eyes were pooled (n = 6, one was excluded due to injury during surgery).
本研究针对干眼病(dry eye disease)小鼠模型中的小鼠角膜上皮细胞开展了单细胞转录谱分析(single cell transcriptional profiling)。整体实验设计如下:我们制备了两类角膜上皮细胞悬液,分别取自对照组与干眼病模型组,实验所用动物为10周龄雄性C57BL/6J小鼠,共7只。参照已发表的实验方案(Shinomiya等,2018),我们摘除了小鼠右侧泪腺(包括眶外泪腺与眶内泪腺),并于泪腺摘除术后第9天处死小鼠。收集左侧对照组眼球共7只进行混合样本制备,右侧干眼病模型组眼球共6只(1只因术中损伤被排除)用于混合样本制备。
