We measured the transcriptome changes in 10-day-old seedlings exposed to Mock and 25% PEG8000 treatment to investigate the differentially regulated genes by ENAP1/2 in response to dehydration Col-0 and enap1-1enap2 were grown in half- strength MS media for 10 days in the 16/8 light period and transferred on filter paper saturated with media solution (Mock) or media containing 25% PEG8000 (Dehydration) for 4h. The whole seedlings were sampled for RNA extraction and RNA-seq libraries.
本研究通过检测经对照(Mock)及25%聚乙二醇8000(PEG8000)处理的10日龄幼苗的转录组变化,旨在探究ENAP1/2在脱水响应过程中调控的差异基因。实验中,将Col-0野生型与enap1-1enap2双突变体幼苗接种于半强度MS培养基,于16小时光照/8小时黑暗的光周期下培养10天;随后将幼苗转移至浸满基础培养基溶液(对照组,Mock)或添加25% PEG8000的培养基(脱水处理组,Dehydration)的滤纸上,持续处理4小时。最后收集全部幼苗样本,用于RNA提取及RNA测序文库的构建。
