Microarray analysis of whole brain RNA collected 48 hr after LPS (4mg/kg i.p.) injection We induced systemic inflammation by injecting mice with LPS. We then performed global gene expression profiling of whole brain RNA and looked for genes that may be involved in behavioural changes. Male C57BL6 mice were injected (i.p.) with 4mg/kg LPS (Escherichia coli serotype 055:B5) or PBS (n=3/group). Mice were euthanised 48 hours after injection and GeneChip Mouse Gene 1.0 ST Arrays (Affymetrix) were used for transcriptional analysis of whole brain RNA.
本研究针对脂多糖(LPS,4mg/kg腹腔注射)造模后48小时采集的全脑RNA开展微阵列分析。实验通过向小鼠腹腔注射LPS构建全身性炎症模型,随后对全脑RNA进行全基因组基因表达谱分析,以筛选可能参与行为改变的相关基因。将雄性C57BL/6小鼠分为两组,分别腹腔注射4mg/kg大肠杆菌055:B5血清型LPS或磷酸盐缓冲液(PBS),每组3只。于注射后48小时处死小鼠,采用Affymetrix GeneChip Mouse Gene 1.0 ST芯片对全脑RNA进行转录组分析。
