To identify the transcriptional signature of human IPCs (hIPCs), we isolated TBR2-positive IPCs from developing human neocortex. We utilized an antibody to mark intranuclear TBR2 in single-nucleus suspensions purified from developing human neocortex procured at the peak of neurogenesis at gestational weeks (GW)14-17, and sorted the TBR2+ nuclei (considered hIPC derived) from the TBR2- nuclei (considered non-hIPC derived). RNA-seq was performed to determine the expression profiles of hIPC-enriched genes.
为鉴定人类IPC(hIPCs)的转录特征,我们从发育中的人类新皮层中分离出TBR2阳性IPC。我们使用抗体标记采自神经发生高峰期(妊娠14-17周,GW14-17)的发育人类新皮层纯化所得的单细胞核悬液中的核内TBR2,并将TBR2阳性细胞核(认定为hIPC来源)与TBR2阴性细胞核(认定为非hIPC来源)进行分选。随后通过RNA测序(RNA-seq)分析,以确定hIPC富集基因的表达谱。
