Choline plays an important role during pregnancy. The study focuses on gene expression profile of whole brain tissues of embryos in mouse female treated with choline deficient diet or chow diet. Overall design: Maternal choline deficiency in mouse embryos. The wild-type female was time-mated with the wild-type male. The treatment group includes normal chow diet (ND) or choline-deficient diet (CDD). Females were treated in different diet since time-mating day. Then 4 embryos per group were collected at E14.5 day old. The brains of embryos were harvested in Trizol for further RNA extraction and samples preparation for RNA-sequencing
胆碱(Choline)在妊娠过程中发挥关键作用。本研究聚焦于饲喂胆碱缺乏日粮或普通日粮的孕鼠胚胎全脑组织的基因表达谱。
实验整体设计:本实验构建小鼠母体胆碱缺乏模型。将野生型雌鼠与野生型雄鼠定时合笼交配,自交配当日起为雌鼠饲喂对应日粮,实验组分为普通日粮组(normal chow diet, ND)与胆碱缺乏日粮组(choline-deficient diet, CDD)。于胚胎发育第14.5天(E14.5),每组收集4枚胚胎,将其脑组织剥离后置于Trizol试剂中,用于后续RNA提取及RNA测序(RNA-sequencing)样本制备。
