Amplicon 16S rRNA gene sequencing was employed to analyse the fine structure, diversity and interactions of microbial biofilms colonizing the outdoor stones of the 19th Century Church of St. John the Evangelist in Melbourne. The hypotheses that bacterial diversity is correlated with building stone lithology, light exposure and surface geometry, respectively, was evaluated. This information may provide the basis for the identification of undesirable deterioration-associated microorganisms and/or the retrieval of beneficial species for preservation of the stone monument.

本研究采用扩增子16S rRNA基因测序(Amplicon 16S rRNA gene sequencing)技术,对墨尔本19世纪圣约翰福音传道者教堂户外石材表面定植的微生物生物膜的精细结构、多样性及种间相互作用展开分析。针对“细菌多样性分别与建筑石材岩性、光照暴露程度及表面几何形态存在相关性”这一假设开展了验证研究。本研究所得信息可为识别与石材劣化相关的有害微生物,以及筛选用于石质文物保护的有益菌种提供理论基础。
