Pten缺失与PI3K抑制可在内皮向造血转化(endothelial to hematopoietic transition)过程中诱导造血干/祖细胞(hematopoietic stem/progenitor cells)发生凋亡;存活的造血干/祖细胞虽可定向分化为所有血液谱系,但干细胞特性(stemness)出现降低。实验整体设计:取36小时受精后(36hpf)的主动脉-性腺-中肾区(AGMs)或5天受精后(5dpf)的尾部造血组织(CHTs),经解剖分离并解离后,通过荧光激活细胞分选(FACS)分离造血干/祖细胞,随后进行单细胞RNA测序。
